As a new revealed game, speculation about Portal 2 will emerge and we got some hints on what to expect.
Speculation
Well we can see from game informer magazine that there is a connection with a wild environment so it can be something that could be related prior Portal events. Portal in a wild environment?! how could that work? Or you continue in a lab test? Also if you've escaped could there be the 'tests' that GlaDOS managed to put us in?

Wants
First thing I want is a longer game. The first one is really great and you can beat it really fast.
It'd be great if we get more dynamic test chambers (if we can call them that). A more dynamic environment.
A connection with Half-Life world. We got it in Portal 1 but something that could create events in HL2 EP3 or a possible HL3.
New kind of enemies. Not saying different but some kind of turret based like the ones that we got.
Updated engine would bring more possibilities on how we use the Portal technology and how do we interact with the environment.
Do NOT Want
I think the game should not destroy what Portal has created. We got a puzzle based game and Portal 2 should continue like that but as the events that occurred maybe a balance between Half-Life and Portal 1 would be a good option.
Portal 2 should maintain the base core/interaction as in Portal 1.
Things that we know
-> Portal 2 is set to drop this upcoming holiday season at full retail price.
-> The return of GlaDOS
-> New characters
-> New areas of the Aperture Science Labs
-> The two-player co-op mode will feature its own campaign with two new playable characters.
