Even when i uncheck it , after i die , i still can't load , and i have to recreate my character

Dead is dead, but I'm pretty sure if you don't check permadead when you make your character you can save without quitting and reload when you die.

I don't think the game autosaves, you probably save manually if you want to continue from a save after dying.

It does - every time you use the stairs, the game autosaves. Saved me quite a bit of work every once in a while.
