Vista fits on a 4.7GB DVD. 16GB is plenty for the OS, not everything.
For everything else you'll use regular hard drives.

Now you may have to plug the flash drives into both eSATA and USB ports since I don't think you'll be able to draw the required power from the SATA port.
So you may have to put like a usb cable from the motherboard inside the case to power the flash drives.
It really seems pretty awesome though for the cheap price. OCZ also has some 16GB and 32GB eSATA flash drives as well if you want to upgrade to some bigger ones.
