Easy--the answer is yes. Don't have the location handy but others can repost from others responses of this sort. I have to run out right now.

what game file do you want to delete?

the save game? or a DS:ToV game file?

to delete the save game you would have to disable steam cloud support for DS:ToV first, then go to:
[path to steam folder]/userdata/[your usernumber]/18050/remote/
and delete "slot1.sav"

to disable steam cloud support:
start steam, click on Library and then on Games. in the game list right-click on "DeathSpank: Thongs of Virtue" and choose "Properties", in the window which should pop up, click on "Updates" and there untick "Enable Steam Cloud..."

however you can just start a new game if you don't like your actual game progress.

if what you want is to uninstall DS:ToV then:
start steam, click on Library and then on Games. in the game list right-click on "DeathSpank: Thongs of Virtue" and choose "Delete Local Content..."

edit: the game files are stored in the "deathspanktov" folder within your steam folder

[path to steam folder]/steamapps/common/deathspanktov/

the default path for the steam folder is:
C:\Program Files\Steam
